川芎嗪和硝酸甘油对体外培养的人血管内皮细胞分泌前列环素的影响

【摘要】 本研究通过体外培养人血管内皮细胞,分别用川芎嗪和硝酸甘油刺激内皮细胞,用放免和血小板聚集试验测定培养液中PGI2的含量及生物学活性。结果表明:川芎嗪具有使内皮细胞分泌PGI2增高的作用,而硝酸甘油则未见有此作用。

【Abstract】 Prostacyclin(PGI2) is the main product of arachidonic acid in all vascular tissue so far tested including human tissue. It is a strong vasodilator of all vascular beds studied including the cerebral circulation.It is also the most potent endogenus inhibitor of platelet aggregation.PGI2 has been tested in the treatment of some vascular diseases. The purpose of this experiment was to find some drugs to stimulate the vascular endothelial cells to synthesis and secrete PGI2.Human umbilical cord vein endothelial cells were cultured in vitro and characterizd by phase contrast microscopy,transmission electron microscopy and immunofluorescence stain with the antiserum against the Ⅷ factor.Three drugs inculding nitroglycerin,ligustrazini hydrochlrioi were added to culture medium.The concentration and biological activities in medium measured comparatively by RIA and platelet aggregation. The results show ligustrazi ni hydrochlrioi could stimulate the vascular endothelial cells to secrete PGI2,but nitroglycrein couldn’t.
