溶于玉米油之α-氰基丙烯酸酯的Ames试验
STUDIES ON THE SOLVENT FOR ADHESIVE OF SPERMATIC DUCT BY AMES TEST
【摘要】 由α-氰基丙烯酸正丁酯和苯酚所组成的输精管粘堵剂是一种新的男性绝育药。该药不溶于水,经气相、红外和核磁等发现DMSO或DMF和粘堵剂混合时,发生化学反应而致粘堵剂结构的改变。乙酸乙酯有明显的杀菌作用,故Ames试验中常用溶剂并不适合作粘堵剂的溶剂。实验发现玉米油易于溶解粘堵剂,且不发生化学反应,可作该药溶剂。在±S9mix条件下,6种阳性剂对TA97、TA98、TA100和TA102均诱发阳性突变,并有剂量—效应关系,但粘堵剂未发现阳性突变反应。
【Abstract】 Adhesive for spermatic duct, which consists of butylcyanoacrylate and phenol, is a new drug mixture for male sterilization.The adhesive is insoluble in water. By gas chromatography, infrared spectrophotometer and NMR spectrometer analysis, it was found that DMSO or DMF and adhesive had chemical action when they were mixed together. Acetic ether had a toxic effect to kill Salmonella typhimurium strain. Therefore, the solvents commonly used in Ames test were not suitable for adhesive. Experiments had shown that corn oil could be a suitable solvent in Ames test. Not only the adhesive was very easily dissolved, but also the adhesive and corn oil together produced no chemical reaction. Under±S9mix condition our results showed that 6 diagnostic mutagens could induce positive reversion mutation and dose-respond curve with TA97, TA98, TA100, TA102 used as new test strain groups.
