大孔硅胶化学键合固定相用于蛋白质分离的反相高效液相色谱

【摘要】 研制不同孔径的硅胶,并进行了C18烷基化学键合。探讨了硅胶孔径对蛋白质的保留、分离和回收率的影响。实验结果表明,硅胶孔径对蛋白原分离的柱效率有重要影响。然而,对本实验中绝大多数蛋白质,在不同孔径柱上,其保留时间基本保持不变。蛋白质的回收率随硅胶孔径增大而有所提高。此外,尚考察了流动相中有机溶剂的浓度对牛血清白蛋白(BSA)保留的影响。

【Abstract】 Silicas with different pore diameters have been prepared and bonded with n-alkyl chain (C18). The influences of pore size on protein retention. resolution and recovery were studied. The results showed that the pore size played a significant role in the column efficiency for protein separation. However, the retention of each protein on the different pore size silica columns was virtually the same for most of the proteins tested. The recoveries of proteins from these columns increased with the pore size. The effect of the concentration of organic solvent in the mobile phase on the capacity of bovine serum albumin was examined.
