成虫石蜡切片抗原免疫荧光技术诊断肺吸虫病

【摘要】 本文初步报告用卫氏肺吸虫成虫石蜡切片抗原通过间接荧光抗体试验(IFAT)检测了血清283份,其中37份痰检虫卵阳性的肺吸虫病人血清,36份(97.3%)呈阳性反应。确诊血吸虫病人血清30份,2例呈阳性(+)。交叉反应阳性率为6.7%。肿瘤病人血清28份及健康人血清188份均呈阴性。这表明成虫石蜡切片IFAT检测肺吸虫病人特异抗体具有较高的敏感性与特异性。并且具有不少优点,成虫固定在福尔马林液中或巳制成切片,在室温内均可贮备较长时间而不失其抗原性,易于保存,制作简易,不需低温的复杂设备,携带使用方便,较目前国内外报告所采用的全虫冰冻切片或虫体碎片固定在玻片上作为抗原的方法为优。因而应用成虫石蜡切片抗原于IFAT可能成为一种更好的,有实用价值的诊断肺吸虫病的新方法。

【Abstract】 The paper reports the prelimmary results of a study on 283 human sera by an indirect fluorescent antibody test (IFAT) with paraffin sections of adult paragonimus westermani as an antigen. Of the 283 sera, 37 collected from paragonimiasis patients with eggs in their sputum were tested, the positive rate being 97.3%; 30 from patients with schistosomiasis japonica, with a cross reaction positive rate of 6.7% while 28 sera from patients with cancer and 188 sera of healthy persons all showed negative results in the tests. It indicates that IFAT with this section antigen has relatively higher sensitivity and specificity in detecting a specific antibody against paragonimiasis. Moreover, the paraffin section antigen has many advantages, such as simple and easy, and in preparation no need of hypothermic equipments. The adult worm either fixed in formalin or made into paraffin sections can be preserved a long time under room temperature without change in its antigenicity, and thus it is portable and ready for use. The paraffin section of the adult worm is believed to be superior to the conventional frozen section or worm particles fixed on microscopic slide in IFAT. Therefore, the paraffin in section of adult worm might be recommended as a preferable technique IFAT for serodiagnosis of oaragonimiasis
