【Abstract】 Thyroid peroxidase is involved inseveral steps of the biosynthesis ofthyroid hormone as in utilization ofH₂O₂:oxidation of iodide to iodi-ne,iodination of thyroglobulin,and a coupling reaction leading toT₄ and T₃ formation.We have measured peroxidaseactivities in 15 specimens of ende-mic goiter and some complication-s,using guaiacol oxidation assayprocedures of Nakashima and Ta-urog （1978）.The method of extra-ction and isolation of TPO was asfollows:after homogenization,centrifugation and solubilization,the activity of the soluble TPOwas measured on spectrophotometerat 470 nm.The results suggest that TPO ac-tivity is various in different stag-es of endemic goiter.In the earlystage,goiter is diffuse.TPO activ-ity is 7 times as high as normaltissue,showing its strong compen-satory activity.When goiter nod-ules occured TPO activity is stillhigher and is 4.5 times as high asnormal tissue.But when the TPOactivity of perinodular tissue wascompared with normal tissue,the-re is no significant difference.Atthe final stage,there is a hugeendemic goiter nodule with degen-erative changes,TPO activity isvery low,showing its compensato-ry deterioration.In endemic goiter with its comp-lications including hyperthyroid-ism,Hashimoto’s disease and car-cinoma,the TPO activity was det-ermined.An autonomous hot nodule was found in a patient with en-demic goiter complicated with hyp-erthyroidism.The TPO activity is5.6 times as high as normal tissue.In a case of Hashimoto’s diseaseenzyme activity is lower than nor-mal.In two cases of carcinoma,enzyme activity is slightlv higherthan normal.更多还原
