诱发玉米抗小斑病突变体的研究 Ⅲ、影响玉米小斑病菌致病毒素作用的因素与生物测定

【摘要】 试验比较了几种致病毒素诱变剂,以病叶冰冻提取液和改良 Fries 培养滤液的效果最理想,高粱米粒培养物的甲醇和蒸馏水提取液效果不理想。为防止致病毒素作用变化、制备诱变剂的小斑病菌最好用病叶保存。用改良 Fries 培养液培养小斑病菌每天振荡六次,每隔1小时一次、每次10分钟。所制备的培养滤液在5℃下可贮存六个月、用高压灭菌器灭菌30分钟及煮沸浓缩均不影响致病毒素的作用。评价致病毒素作用以比较抑制种子根生长的种苗萌发包法效果最好。愈伤组织无性系和试管苗抗病性的测试可用定量孢子的纸片法。

【Abstract】 Several Hm-pathotoxin inductors were comparied.The extracts of free- zed diseased leaves and filtrates of improved Fries medium cultures were best while the methyl-alcohol and distilled water extracts of sorghum grain cultures were not so good.In order to prevent the role of Hm-patho- otoxin from being changing.It is better to preserve the isolstes of Hclmint- hozporiyn.mnydis to be used in induction studies in diseased leaves when improved Fries medium was used to cutture the Helminthosporium maydis it should be shaken six times each day(1 hr interval 10min/time).The cul- tural filtrates of Helminthosporium muydis could be maintained six months at 5℃,and the role of Hm-pathotoxin was not affected after sterilizing 30 minutes in autoclave(121C,151bs)or boiling for concentraction.The best method to estimate tile role of Hm-pathotoxin was to germinatethe seeds and compared the growth of primary roots(?)The disease resistance of the pollen callus tissue and tuber-seedling could be tested by the method of fragment-paper with quantified conidia of the pathogen.
