人类白内障晶状体的超微结构和元素分析

【摘要】 本文报道人成熟型白内障晶状体的超徽结构和元素分析。作者使用附有能谱仪的JSM-35C扫描电镜对晶状体纤维细胞的巯、磷、钙进行微区分析。结果表明,硫的谱峰很高,磷和钙的谱峰则很低。此外,线扫描分析证明,沿扫描线硫在晶状体纤维细胞内的分布比较均一。通过扫描电镜观察,发现其纤维细胞外形不规则,球-和-凹的连接不易看到。在不同区域的晶状体纤维细胞出现大量破坏及球体的形成。而最明显的改变是在品状体局部纤维细胞的扁平面形成无数刺状突起,这些突起具有不同的大小和形态,分布也不均匀。

【Abstract】 In this paper, we report the ultrastructure and elemental analysis of the mature human cataractons lens fibers. All the lenses were obtained from cataract surgery. Immediately after operation, the lenses were fixed with 3% glutaraldehyde buffered with 0.1 M eacodylate (pH 7.2) for 48 hr at 4℃. The well-fixed lens was cut into two halves: one half was used for conventional scanning electron microscopy and the other half for elemental analysis. For the detection of the elements such as sulfur, calcium and phosphorus in the mature human eataractous lenses, a JSM-35C scanning electron microscope equipped with an Energy Dispersive X-ray Analyzer was used.Results from this study demonstrate that the resultant spectrum shows a higher peak at the sulfur energy level and a relatively small peak for calcium and phosphorus. In addition, line-scan analysis of this lens shows that the sulfur was rather uniformly distributed along the scan line.In the present study, the eataractous lens fiber cells are irregular in size and shape and their ball-and-socket junctions previously demonstrated are not readily identified throughout the length of these cells. The cavernous structure of lens fiber is formed on the surface of the cell. It appears to be formed by the rupture of the cell membrane. The massive disruption of fiber cells and formation of globules are also noted in various zones of the lens. The most significant change observed in the human eataractous lens is the numerous thorn-like protrusions of various sizes and shapes on the surface of the fiber cells. These kind of cells are located in various zones of the lens.
