【Abstract】 This paper describes splinocytes from Balb/c mice immunized with HBsAg and fused with SP2/0 myloma cells under the action of PEG to yield two hybridomas (SH1D9, SG3B10). Hybrids of SH1D9 and SG3B10 after long-term culture in vitro produced monoclonal antibodies with titers of 1:10000 and 1:8000 respectively by RIA and ELISA, The monoclonal antibody secret- ed by SH1D9 was anti-HBs/a, while that secrected by SG3B10 was anti-HBs/y. All of them belong to the mouse IgG subgroup. Purified McAb/a, y and a+y were used for coating sheep RBC and labelling with 125I to make RPHA-SRBC and RIA reagents respectively. McAb SRBC and 125I-McAb were used to test HBsAg in comparison with PcAb-SRBC and 125I-PcAb. The resulds demonstrated that the monoclonal antibodies havehigh specificity to HBsAg subtype determinant and can be used as a diagnostic reagent to test single antigenic determinants. McAb/y may be useful for the study of HBA/y subtype infection.更多还原
