麦类赤霉病穗腐镰刀菌酯酶同工酶谱的聚丙烯酰胺凝胶电泳分析

【摘要】 采用聚丙烯酰胺凝胶电泳技术,对23个麦类赤霉病穗腐镰刀菌菌株(分属于16个种和变种)的正向极酯酶同工酶进行了分析。并研究了影响测定的一些因素。从1980~1982年,经多次测定,得到了稳定的结果。这些镰刀菌种和变种间的酯酶谱有明显而稳定的特异性。7个来源或性状不同的禾谷镰刀菌菌株的酯酶谱,彼此间有一些差异,但也有共同的酶带。因此把酯酶用作它们分类的辅助手段是有意义的。在本试验中,盘状凝胶电泳较薄层水平板凝胶电泳的分离效果好。10%的凝胶浓度优于7%的浓度。凝胶用过硫酸铵聚合,对所测菌的正向极酯酶,不造成失活。所测试的培养温度和日期对酶谱无重大影响,而培养基种类则有一定影响。

【Abstract】 The esterase isozymes of the Fusarium sp.causing headblight of whe-at and barley were studied by polyacrylamide gelelectrophoresis.Twen-ty four fungal isolates representing 17 species and varieties of the ge-nus Fusarium were used in this study.They were F.graminiarum,F.cul-morum,F.moniliforme,F.avenaceum,F.avenaceum var.herarum,F.ni-vale,F.acuminatum,F.sulphureum,F.equiseti,F.eqiseti var.longipes,F.poae,F.camptoceras,F.semitectum,F.concolor,F.fusarieides,F.oxysporum,F.oxysporum var.vasinfectum.After many repetitions,the stable resultes were obtainod.The ea-terase patterns of different Fusarium sp.showed distinetly differencesbetween them could be used as a useful taxonomic criteria.The este-rase patterns of 7 isolates of F.graminiarum collected from different ge-ographic localities appeared a little difference between them,but therewere some identical bands of esterase isozymes in their esterase patterns.Disc polyacrylamide gel electrophoresis and 10% gel were better thanhorizontal thin-layer polyacrylamide gel electrophoresis and 7% gelfor eleetrophoretic separations of the esterase isozyms of Fusarium sp.The cultural daya(range from 4 to 14 days)and the temperature(rangefrom 15 to 30℃)did not influenc the patterns,but the mediums influen-ced them to a certain extent.
