【摘要】 本文利用Sephadex LH20凝胶过滤法分离结合和游离的甾体,以定量人子宫内膜胞浆中的孕酮受体(PRc),并同时采用葡聚糖-活性炭吸附法(DCC)对标本进行配对测定。这两种方法测得的特异性结合量(Bs)以及总结合比率(Bt/T)之间,经统计学处理显示有密切正相关存在。LH20法是一种有效的和较为温和的分离测定受体的方法。由于LH20法能直接测定游离甾体浓度,故有利于回收率的精确测定。结果表明,采用LH20法测得的PRc的结合参数可能更为可靠。更多还原

【Abstract】 Method using gel filtration on Sephadex LH20 to seperate bound and free steroid wasstudied for quantifying progesterone receptors in the cytosol of human endometrial cells.Samples were also assayed by using dextran-coated charcoal adsorbent technique. Compari-son of the binding parameters obtained using LH20 with those using dextron-charcoal methodsshowed good agreement in the estimation of specific binding capacity and bound/total ratio.Specific binding capicity was consistently higher in the LH20 method which was more efficientand mild and at the same time, the free steroid concentration and the recovery rate could bemeasured directly. It seems likely that the set of values obtained with LH20 is probablymore reliable.更多还原
