人参茎叶皂甙拟促皮质激素样作用的研究

【摘要】 本文报告了人参茎叶皂(以下简称CS)可增加正常大鼠肾上腺细胞内cAMP的含量,且与ACTH有协同作用。去垂体的大鼠则无此作用说明GS的拟ACTH样作用是通过垂体,而非直接作用于肾上腺皮质。同时观察了人参茎叶二醇组皂甙、三醇组皂及人参单体皂Rb2、Re、Rg、Rd对肾上腺细胞内cAMP含量的动态变化,以二醇组皂及单体Rd作用最显著,故二醇组皂和单体皂Rd可能是兴奋垂体的主要皂。本实验比较了GS和人参皂对垂体的兴奋作用,两者作用强度基本相同,故在此方面GS可代替人参皂。这为人参的综合利用提供一定的实验依据。

【Abstract】 Effects of total ginsenoside, isolated panaxadiol and panaxatrial from the stems and leaves of panax ginseng on adrenal intracellular cAMP level in rats were demonstrated with competitive protein-binding technique.When total ginsenoside was administered to rats intraperitoneally,adrenal intracellular cAMP level was increased, most significantly 30 min after the injection. The isolated panaxadiol and panaxatrial also increased the adrenal intracellular cAMP level,but the effect pro- duccd by panaxatrial had no statistical significance.In rats which had received hypophysectomy before the injection, no increase in adrenal intraccllular cAMP level was observed. When ACTH was given shortly after the ginsenosidcs, the increase in adrenal intraeellular cAMP level was much more enchanced. In addition, although ginsenoside Rb2. Rg and Re did not increase the cAMP level in the adrenal cortex, ginsenoside Rd did cause a marked increase. Thus, our study indicates that ginseng saponins may act on the hypophysis primarily and stimulate ACTH secretion, resulting in an increase in the level of cAMP in the adrenal cortex. Ginsenoside Rd seems to be one of the chief ginsenosidcs that act on the hypophysis.
