流行性出血热病原学研究——Ⅰ、用非疫区黑线姬鼠分离病毒及其特异性检查
Etiological Studies of Epidemic Hemorrhagic FeverI. Virus Isolation in Apodemus agrarius from Non-endemic Area and Its Antigenic Characterization
【摘要】 在用非疫区黑线姬鼠分离EHF病毒的传代中,发现感染鼠鼠肺中存在一种可传代的EHF相关因子。用间接免疫荧光法对其特异性作了检查:101份EHF患者血清96%阳性,175份正常人及其他疾病患者血清全部阴性;20例EHF患者双份血清,18例的恢复期有4倍以上抗体滴度升高;12例EHF患者血清对KHF抗原及EHF相关因子的阴、阳性反应基本一致;抗呼肠孤病毒血清和抗类呼肠孤病毒血清等全部阴性;血清编码双盲法检查和预期结果一致;抗KHF血清对EHF患者血清的免疫荧光反应有显著阻断作用,抗呼肠孤病毒Ⅰ~Ⅲ型多价血清无此作用。上述结果可初步确定,此相关因子是流行性出血热病毒。
【Abstract】 A transmissible EHF related agent was discovered in lung tissue of infected Apodemus agrarius in the course of serial passages for EHF virus isolation in Apodemus agrarius from non-endemic area. Antigenic specificity of the isolates was detected by indirect fluorescent antibody technique ( IFAT ):among 101 convalescent sera of EHF patients tested, 96% were positive; 175 sera of normal persons and patients suffering from other diseases were all negative; out of 20 paired sera of EHF patients, 18 showed 4 fold or more rise of antibody titers; 12 sera of EHF patients tested with KHF antigen previously were reexamined with EHF antigen actually with the same positive or negative results; tests made with antisera against reo-virus (type 1) and reovirus-like agent (orbivirus?) gave consistently negative results; blind tests of coded sera of EHF patients and normal persons produced similar expected results; obvious blocking effect of KHF antiserum to immuno-fluorescence elicited by EHF patient’s serum was demonstrated, but no such effect was found with polyvalent antisera of reovirus types 1-3. The above results demonstrated preliminarily that the isolated EHF related agent was the causative agent of EHF.
