【摘要】 本文报道PLC/PRF/5细胞培养液经羊-抗 HBs 亲和层析柱部分纯化后,可获得HBsAg。免疫电镜检查显示以20毫微米球形颗粒为主。用以免疫5只小白鼠和2只豚鼠后,分别有4只和2只产生特异性抗-HBs。用免疫豚鼠血清提取的 IgG,致敏醛化羊红细胞做反向被动血凝试验,可被特异性 HBsAg 所抑制,包被聚苯乙烯板做固相放射免疫测定,可出现阳性反应.作者认为提高 PLC/PRF/5细胞的 HBsAg 产量,可能取代血液来源的 HBsAg,用于血清学诊断。更多还原

【Abstract】 For studies on immunogenicity of HBsAg produced by PLC/PRF/5 cell line,cul-ture media were collected and partially purified by means of sheep anti-HBs affinity chromatography.After being concentrated,partially purified HBsAg was examined un-der immuno-electronmicroscope and chiefly 20nm spherical particles were observed.These were used to immunize the mice and guinea pigs together with Freund completeadjuvant.Four out of five mice and all two guinea pigs produced anti-HBs,whichcould be specifically inhibited by HBsAg.IgG was fractionated from guinea pig anti-HBs and was used to coat aldehyde treated red blood cells and microtiter plate wellsfor reverse passive hemagglutination and solid phase radioimmunoassay respectively.Both tests showed positive results but were less sensitive in comparison with highlyimmunized antiserum.It is suggested that by increasing the amount of HBsAg pro-duced by PLC/PRF/5 in the future,it may be possible to substitute the present HBsAgpurified from human serum for use in diagnosis.更多还原
