低共熔溶剂提取丹霞铁皮石斛多糖工艺优化及其抗氧化活性评价

【摘要】 采用低共熔溶剂提取丹霞铁皮石斛多糖,考察液料比、提取温度和提取时间对丹霞铁皮石斛多糖提取效率的影响,并对提取工艺参数进行优化,最后测定丹霞铁皮石斛提取液的抗氧化能力。结果表明,当液料比80∶1(mL/g)、提取温度80℃、提取时间1.5 h时,丹霞铁皮石斛多糖得率为404.4 mg/g。当多糖浓度为1.566 mg/mL时,丹霞铁皮石斛低共熔溶剂提取液对DPPH自由基、超氧阴离子自由基和羟基自由基的清除率分别为59.64%、55.61%和70.70%。综上,氯化胆碱/乳酸低共熔溶剂提取法具有制备高得率、较高抗氧化活性丹霞铁皮石斛多糖的潜力。

【Abstract】 Provincial Key Lab of Utilization and Conservation of Food and Medicinal Resources in Northern Region,Abstract:In this study,the polysaccharides from Danxia Dendrobium officinale was extracted by deep eutectic solvent. The effects of liquid-to-solid ratio,extraction temperature,and extraction time on the extraction efficiency of polysaccharides from Danxia Dendrobium officinale were investigated. In addition,the extraction process parameters were optimized. Finally,the antioxidant activity of Danxia Dendrobium officinale extracts was determined. The results showed that when the liquid-to-solid ratio was 80∶1(mL/g). The extraction temperature was 80 ℃,and the extraction time was 1.5 h,the yield of polysaccharides from Danxia Dendrobium officinale was 404.4 mg/g. The scavenging rate of the Danxia Dendrobium officinale extracts by deep eutectic solvent for DPPH radicals,superoxide anion radicals,and hydroxyl radicals was 59.64%,55.61%,and70.70%,respectively,when the polysaccharide concentration was 1.566 mg/mL. This study indicated that the choline chloride/lactic acid deep eutectic solvent extraction method had the potential to prepare polysaccharides from Danxia Dendrobium officinale with high yield and high antioxidant activity.
