虾夷马粪海胆“红斑病”病原弧菌特性及致病性
Biological characteristic and pathogenicity of the pathogenic vibrio on the "redspot disease" of Strongylocentrotos intermedius
【摘要】 从患“红斑病”的虾夷马粪海胆病灶处分离出一种可疑病原菌,编号为0205。经感染实验证实,此菌可通过体表创面感染海胆,引起“红斑”症状出现。发病与水温关系密切,高温易发病,低温发病率降低。0205菌革兰氏阴性无芽孢,短杆近球形,具单极毛,生长需盐。对其进行系统鉴定归为弧菌属,其生理生化特性与伯捷氏细菌系统第9版中所列模式种均不相同。经16S rRNA gene序列分析及同源性检索发现该菌与GenBank中AJ002566菌同源性为99.8%,AJ002566菌由德国学者Yakimov直接提交其16S rRNA gene序列1~1476bp,无分类位置。
【Abstract】 A kind of red spot disease burst out in cultured sea urchin Strongylocentrotos intermedius at Dalian coastal area in the summer 2002.Typical symptom are spots with wine colored mucous substances on the surfaces of diseased sea urchins.The inside substances of diseased sea urchins outflowed by a hole of spotting lesions in serious condition.The disease broke out in short time and induced a mass mortality in summer.Nevertheless,the disease occurred rarely in autumn and the death of sea urchins were also observed singularly.A total of two stains were isolated from the mucus on red spot of the diseased samples,numbered 0205 and 0206.Experimental infections were performed by the immersion method in five different concentrations of bacterial suspension ranged from 103 cell·mL-1 to 108 cell·mL-1 at 23 ℃ and 14 ℃ for 2 h.The only wounded sea urchins who lost their spins challenged with 0205 occurred symptoms the same as natural disease and the infection rate of each concentration was a hundred percent except the concentration 103 cell mL-1.So the stain 0205 was considered as the causative bacteria.The stain 0205 revealed gram negative,without endospore,curved rods or short rods resembling coccus,with a single polar flagellum,salt requirement.Some tested items of morphological,physiological and biochemical characteristics do not completely match to all of the species modes which belong to Vibrio genus in Bergy’s Manual of Systematic Bacteriology.The 16S rRNA genes of 0205 were sequenced and compared with the database information in Genbank.The 16S rRNA genes sequence of 0205 exhibited the highest similarity to the 16S rRNA gene sequence(1-1 476 bp) of AJ002566 directly offered by Yakimov.
