【摘要】 目的 :探讨蛭参合剂延缓 5 / 6肾切除大鼠肾小球硬化的作用。方法 :随机将动物分为空白对照组 ,5 / 6肾切除大鼠模型对照组和蛭参合剂组。分别于手术后 4周、8周检测各组大鼠血红蛋白、血肌酐、血尿素氮和 2 4h尿蛋白含量 ;Masson和免疫组化染色结合图像分析系统半定量观察肾小球内胶原纤维和转化生长因子 β1(TGF - β1)、纤维连接蛋白 (FN)及Ⅲ型胶原 (CollⅢ )的积分光密度 (IOD)。结果 :与模型组比较 ,蛭参合剂组可减少血肌酐上升程度(P <0 .0 5 ) ,降低尿素氮 (P <0 .0 5 )和 2 4h尿蛋白量 (P <0 .0 5 )。并随着时间延长而显示出延缓了血肌酐上升速度的趋势 (P >0 .0 5 )。肾小球内TGF - β1、FN、CollⅢ和胶原纤维的病理性聚集的减少具有明显差异 (P <0 .0 1)。结论 :蛭参合剂能延缓大鼠肾小球硬化和慢性肾衰竭进展速度。更多还原

【Abstract】 Objective:To investigate the effects of preventing progress of glomerular sclerosis by the Zhi-Shen mixture in rat with 5/6 nephrectomy. Methods:The rats were randomly assigned to Zhi-Shen mixture treatment group ,control group and normal group. Hemoglobin, serum creatinine(Scr),blood urea nitrogen(BUN) and 24 h urinary protein were measured at 4th and 8th week after the nephrectomy. Renal pathology changes were evaluated at the same time. The renal tissues were stained by immunohistochemical and Masson methods. Then integra optical densit(IOD) of Collagen fibrin,TGF-β 1, FN and Collagen Ⅲ(Coll Ⅲ) of the glomerular were semi-quantified by image analysis system.Results:In comparison with the control group,Scr?BUN and the 24 h urinary protein were decreased in Zhi-Shen mixture group(P<0.05).The immunohistochemical result showed that Collagen fibrin ,TGF-β 1,FN and Col Ⅲ of the glomerular were significantly decreased in Zhi-Shen mixture group(P<0.01).Conclusion:This Zhi-Shen mixture can delay the progress of glomerular sclerosis on 5/6 nephrectomized rats.更多还原
