用同步辐射X荧光分析法研究兔肝金属硫蛋白中的微量元素
Study on trace elements distribution in rabbit metallothioneins by synchrotron radiation X-ray fluorescence analysis
【摘要】 采用同步辐射X荧光分析法 (SRXRF)直接测定了兔肝金属硫蛋白 (MT)中的金属离子含量 ,以及经过十二烷基硫酸钠 -聚丙烯酰胺凝胶电泳 (SDS -PAGE)分离后凝胶板中蛋白条带上金属离子的分布。实验结果表明 ,兔肝MT - 1以含锌为主 ,占总金属含量的 97.9% ;兔肝MT - 2以含锌、镉为主。本工作表明 ,采用SRXRF技术可实现MT电泳分离后蛋白条带内金属离子分布的原位测定
【Abstract】 The distributions of metal ions in rabbit metallothionein (MT) and its protein band in sodium dodecyl sulfate-polyacrylamide gel after electrophoresis were determined by synchrotron radiation X-ray fluorescence (SRXRF) analysis. Results showed that MT-1 mostly contained zinc, which amounted to 97.9% of the total metallic content and MT-2 mainly contained zinc and cadmium. This paper also introduced the method to determine the relative amounts of protein-bound metal ions in situ by combination of electrophoretic separation and SRXRF analysis.
