丙泊酚对兔高氧性肺损伤保护作用的实验研究

【摘要】 目的:探讨丙泊酚对高氧性肺损伤的保护作用及可能的机制。方法:24只成年兔随机分为空气组(A组,吸入21%O2),高氧组(B组,吸入95%O2)和丙泊酚组(C组,吸入95%O2并输注丙泊酚),每组8只。实验至4h时处死各组动物,取肺组织制成匀浆和低温差速离心法制备肺线粒体,比色法测定肺组织匀浆中一氧化氮(NO)、丙二醛(MDA)、还原型谷胱甘肽(GSH)含量;氧电极法测定兔肺线粒体呼吸功能;电镜下观察肺泡Ⅱ型上皮细胞线粒体超微结构。结果:B组Ⅲ态耗氧速率(R3)和呼吸控制率(RCR)显著低于A组(P<0.01),C组R3和RCR较A组降低但无显著性;B组肺组织匀浆中MDA和NO含量较A组显著升高(P<0.01),C组显著降低(P<0。01),B组GSH降低而C组GSH升高(P<0.01);电镜下B组肺泡Ⅱ型上皮细胞内线粒体明显肿胀,C组线粒体肿胀较轻。结论:丙泊酚抑制高氧引起的肺组织脂质过氧化反应和氧自由基生成,减轻线粒体结构和呼吸功能损害。

【Abstract】 Objective: To investigate protective effects of propofol on the lung injury induced by 95 % hyperoxia inhalation in rabbits. Methods: Twenty-four adult rabbits without aneathesia were divided randomly into three groups: control ( groupA) , hyeproxic (group B) and propofol (group C) ,eight in each group. All animals were sacrificed after 4 hours of experiment and lungs were removed for homogenization. To obtain a purified lung mitochondria! fraction by centrifugalization, mitochondrial oxygen uptake were measured with an oxygen electrode; the content of nitric oxide (NO) , malondialdebyde (MDA) , superoxide dismutase(SOD) and reduced glutathione(GSH) were measured in lung homogenates with chromom-etry; the morphological changes of mitochondria in alveolar type II cells with transmission electron microscopy were observed . Results: Compared with group A, oxygen uptake of state HI and RCR of lung mitochondria in group B reduceded markedly( P < 0.01 ) ,but lower in group C with no significant;the levels of MDA and NO rose in group B significantly ( P < 0.01 ) but declined in group C significantly compared with group B ( P < 0.01 ) ;the levels of GSH were lower in group B but higher in group C than in group A ( P < 0.01 ); mitocondrias were all swelled markedly in group B and group C, but lighter swelling was observed in group C than that in group B. Conclusion: 4 hours of 95 % hyperoxia inhalation result in respiratory function and morphology of lung mitochondrias damaged,with lipid peroxidation process enhanced in rabbit lungs.Propofol infusion can provide protection for hyperoxia-induced lung injury to some extent.
