【摘要】 本文综合并改进一些常用的分析技术,设计出一套较全面地检测尿激酶粗品质量的快速简便方法。运用本法测定了贵阳市中西工业公司生产的尿激酶粗品的酶活力、蛋白质含量、比活力;并按本法将粗品分离纯化,测定其中高、低分子量尿激酶的相对含量。经与有关资料比较,证明本方法是可靠的,也证明该公司产品在上述质量指标方面是合格的。更多还原

【Abstract】 This paper reports the results of quality exam of semifinished urokinase produced by Guiyang Zong-Xi Industry Company with the methods of synthetic substrate and UV absorbance. The results are: Activity 34000-36000 IU/g, protein contents 68-110mg/g, and specific activity 327-525 IU/mg·protein. In order to check the factional composition, the semifinished urokinase were purified by Sephadex G-25 column chromatography follewed by D256 Ion-exchange resin and CM-Cellulose colunn chromatography.Two forms of urokinase with molecular weight of 55000 and 34000 were separated by SDS-PAGE. Quantitative analysis showed that the high molecular weight urokinase accounts for 56% of total value and low molecular weight urokinase accounts for 44%.更多还原
