猫备用背根节内NT-3和相关神经肽的表达及其作用探讨

【摘要】 在胚胎早期神经营养素-3(neurotrophin-3,NT-3)对神经元的存活、突起生长及靶选择发挥着重要的作用,随着胚胎的发育其表达逐渐减少,功能作用也随之减弱。在成年动物神经损伤后,NT-3的表达再次增高,参与到损伤修复的过程中。有实验显示成年猫部分背根损伤后备用背根节中NT-3mRNA表达上调,同时保留的备用背根在脊髓内发生了侧支出芽,以代偿丢失的初级传入纤维,提示增多的NT-3很可能参与了上述的代偿、修复过程。 为了探明成年猫正常背根节和部分去背根后备用背根节内源性NT-3对节内神经元的作用和作用模式,NT-3对备用背根侧支出芽的影响。以及背根节内与NT-3功能可能有相关关系的神经肽的表达及作用,本文采用CTB神经束路追踪、分离神经元培养、免疫组织化学、免疫荧光、酶联免疫吸附等技术从三个方面进行研究:NT-3促背根节神经突起生长作用探讨;备用背根节内NT-3表达的变化及作用模式探讨:背根节神经元内NT-3与肽类物质的共存及相关作用的探讨。

【Abstract】 Neurotrophin-3 (NT-3) plays an important role for neurons survival, neurites growth and target selection in the early embryonic development. Accompanying embryonic development NT-3 down-expressed gradually. When nerve injury NT-3 up-regulated again, which is associated with the repairing process. After partial deafferentation of spinal cord of adult cat, NT-3mRNA in spared dorsal root ganglion (DRG) up-regulated, meanwhile, the spared dorsal root collateral sprouting occurred in the spinal cord to replace the lost primary afferent nerve fibers. It suggests that increased endogenous NT-3 from spared DRG may promote this process.In order to explore the effect and mechanism of endogenous NT-3 from normal and spared DRG; the influence of NT-3 on spared dorsal root collateral sprouting; and expression and functions of some neuropeptides which are related to NT-3, this study was designed to detect the collateral sprouting of spared dorsal root; the effect of endogenous NT-3 from the spared DRG on ganglionic neurons; the percentage of NT-3 and TrkC positive neuron of lumbar DRG of adult cat; the expressional change of NT-3 in the spared DRG; the co-localization of NT-3 and NPY, CGRP, VIP, Galanin in the adult lumbar DRG respectively; the expressional change of these neuropeptides in the spared DRG, using the techniques of CTB neural-tracing, cell culture, immunohistochemistry, immunofluorescence and ELISA methods.
