EGF、雌二醇和生长抑素对人肝癌细胞生物学行为影响的研究

【摘要】 目的 原发性肝癌是我国、也是世界上最常见的恶性肿瘤之一,严重危害人类的健康。自20世纪90年代以来,我国肝癌的死亡率已上升到肿瘤死亡率的第二位。近年我国肝癌治疗效果得到明显提高,但由于肝癌发生机制尚未阐明,治疗效果仍不满意。肝癌发生是一个多因素、多步骤的复杂生物学过程。HBV、HCV、表皮生长因子(EGF)、性激素、细胞因子、癌基因及抑癌基因等多种因素参与并促进了肿瘤的发生。EGF促进肝癌细胞DNA的合成,促进肝癌的发生与发展。EGFR广泛分布于肝细胞及肿瘤细胞,与细胞增殖、分化调节、肿瘤转化密切相关。EGF和EGFR在肝癌存在过表达,与肝癌的形成关系密切。雌二醇(E2)主要在肝脏代谢和灭活,可以诱导肝原癌基因活化。雌激素受体(ER)调控基因表达,与肝癌发生、发展和预后有关。生长抑素(SST)和生长抑素受体(SSTR)在体内分布广泛,SST及其类似物(SSTA)在体外可以抑制多种人肝癌细胞株生长,可以分别对EGF、EGFR、E2、ER产生抑制作用,从而发挥抗肿瘤作用。在EGF、E2、SST及其受体的信号传导途径中,有丝分裂原活化蛋白激酶(MAPK)发挥了重要作用,成为它们之间相互作用的基础。EGF、E2、SST及其受体与人肝癌细胞增殖的关系及其相互作用机制尚未阐明,且缺少系统研究的报道。本课题试图通过研究人重组表皮生长因子(rhEGF)、人β-雌二醇(β-E2)和人生长抑素(hSST)对人肝癌HepG2细胞增殖的影响和HepG2细胞EGF、EGFR、E2、ER、SST和SSTR的变化,探讨上述诸因素在肝癌发生中的相互作用及其机制,为应用生长抑素治疗肝癌提供实验及理论基础。
【Abstract】 Objectives Primary liver cancer(PLC) is one of the commonest malignant tumors in China and the world,which damages the health of the mankind.The death rates of liver cancer have ranked the second among the tumor death rates in China since 1990.In recent years the therapeutic effects of PLC have been improved greatly in our country.Incomplete understanding of the carcinogenesis of PLC leads to the unsatisfactory therapeutic effects. The formation of PLC is a very complicated, multistep,biological process initiated by the pathogens. Hepatitis B Virus, Hepatitis CVirus, epidermal growth factor(EGF), sex hormones,cytokines,oncogenesand tumor suppressor genes etc. play very important roles in the carcinogenesis of PLC.EGF stimulates DNA synthesis of liver cancer cells and promotes the progress of liver cancer.EGFR,relating to the cellular proliferation,differentiation and transformation, appears on hepatocytes and many kinds of cancer cells.The overexpressions of EGF and EGFR were found in liver cancer indicating a close relationship with liver cancer formation.Estradiol(E2) is metabolized and deactivated mainly in the liver.E2 may activate the hepatic proto-oncogenes.Estrogen receptor(ER) regulates gene expression and plays a role in the formation,advance and prognosis of liver cancer.
