螺旋藻多糖的研究

【摘要】 螺旋藻多糖是一种水溶性多糖(Polysaccharide of spirulina platensis, PSP),有着许多独特的生理功能和疗效,有利于提高机体的免疫能力,市场需求量与日俱增。利用混合培养技术和其它生物物理方法,能有效地提高螺旋藻的生长速率和生物产量,并从中获取更多的有效成分,如藻蓝蛋白和多糖,已成为当前研究开发螺旋藻的热点之一。本论文系统地研究了钝顶螺旋藻优化培养的条件和方法以及水溶性多糖的分离、提取、纯化方法,并对钝顶螺旋藻水溶性多糖的理化性质、化学结构及抗自由基活性进行了较为全面的研究和探讨。主要研究内容和结果如下: 1.钝顶螺旋藻多糖的优化培养技术 (1) 从提高螺旋藻的生长速率,生物产量及多糖含量的目的出发,研究探讨了温度、pH、氮源浓度及钙离子对螺旋藻生长及多糖含量的影响。研究表明:在30~35℃范围内,螺旋藻混合营养生长良好,最终收获的细胞干重、蛋白质和多糖的总量最多。pH对螺旋藻生长的影响不受低浓度葡萄糖(1.0g/L)存在的影响,其最适pn与光合自养生长的最适pH8.5~9.5相同。氮源不足或过多的氮源都降低了螺旋藻多糖的合成。2.0 g/L的硝酸钠是支持钝顶螺旋藻混合营养生长及提高生物产量和多糖含量的最适浓度。钙离子对钝顶螺旋藻的生长和多糖的合成有明显的影响,0.04 g/L的可溶性氯化钙是利于钝顶螺旋藻生长和提高多糖含量的最佳浓度。 (2) 光强和光质对钝顶螺旋藻的光合自养生长和多糖的合成有明显影响。当光强从2klux上升到6klux时,比生长速率增加,多糖含量也随之增加。不同光质处理中,红光有利于螺旋藻的生长和干物质的积累,但蛋白质含量和多糖含量不是最高,绿光不利于钝顶螺旋藻的光合自养生长,但其蛋白质含量和多糖含量却较高。 (3) 一定的磁场强度(0.1~0.6T)能促进钝顶螺旋藻的生长和多糖的产量的提高。

【Abstract】 Polysaccharide of Spriulina Platensis (PSP) is , a water-soluble polysacchride advavtageous to enhance immunological competence which have many specical physiological accommodation function and medical effects .In recent years, there is a great market demand for PSP. Utilization of mixotrophic culture and other biophysical method can enhance the growth rate and cell yield of Spirulina Platensis .It is become much more popular to how to obtain much more bioactive compositions such as phycobiliprotein and polysaccharide. This paper has made comparatively through exploraton on the optimal culture conditions and methods, sparation and purification of Polysaccharide of Spirulina Platensis (PSP), the physical and chemical properties of this polysaccharide and its antioxidation was also deterrmined.The main aspects and results are as follows:(1) The effects of temperature, pH, nitrogen concentration and Ca2+ on the celluar growth of S.platensis and the polysaccharide content were studied and discussed in order to enhance the specific growth rate, the cell yield of S. platensis and the content of PSP. Those results showed : the mixotrophic growth was better and obtained finally much more cell dry-weight, much more total content of protein and polysaccharide from S. platensis during the temperature of 30 ℃ 35 ℃. In mixtrophic culture , the effects of pH on the growth was not affected by the low concentration of glucose (1.0g/L), its suitable pH was the same as suitable pH of photoautophic growth. The deficent or supernumerary nitrogn was all advantageous to biosynthesis of PSP. The concentration of 2.0g/L NaNO3 gives the highest specific growth rate , enhance of cell yield and the contents of PSP. Ca2+ have an obvious effect on the growth of S.platensis and the synthesis of PSP .water-soluble CaCl2 of 0.04g/L is the suitable concentration for S.platensis to enhance the specific growth rate and the content of PSP.
