CD147/Basigin、TAP63/△ NP63与眼睑基底细胞癌发病机制的相关研究

【摘要】 目的:研究CD147在眼睑基底细胞癌(BCC)中的表达状况及其与肿瘤预后的相关性;研究眼睑BCC中MMP2的表达,分析其生成与CD147表达的关系;研究ΔNP63/TAP63在眼睑BCC和表皮中的表达,以揭示其在表皮更替和眼睑BCC发生中的分子机理。 方法:运用荧光定量PCR方法检测CD147在25例眼睑BCC中的基因水平,用激光扫描共聚焦免疫荧光方法检测其蛋白表达状况,研究其与肿瘤复发、组织病理学分型等预后相关指标的关系;用免疫组织化学方法检测MMP2的表达状况,分析肿瘤细胞和周围基质细胞中MMP2的生成与CD147表达的相关性;用荧光定量PCR方法检测眼睑BCC中ΔNP63/TAP63的基因水平,研究其在肿瘤发生中的分子机理;用荧光定量巢式PCR方法检测25例正常表皮中不同类型细胞中TAP63/ΔNP63的表达状况,研究二者在表皮正常更替中的作用。 结果:(1)20例眼睑BCC组织和6例正常眼睑皮肤组织中检测到CD147的RNA转录,正常皮肤组的相对拷贝数远低于眼睑BCC各组。BCC未分化原发组与BCC未分化复发组间差异具统计学意义。免疫荧光结果与PCR相一致。(2)MMP2在眼睑BCC分化组、未分化原发组和未分化复发组中的阳性表达率分别为40%、47.06%和100%,且与肿瘤复发相关。(3)CD147的表达与肿瘤周围基质细胞MMP2的表达相关,而与肿瘤细胞中MMP2的
【Abstract】 Objectives: To investigate the expression of CD147/basagin in eyelid basal cell carcinoma(BCC) and the correlation between its expression and prognosis. To study the expression of MMP2 in eyelid BCC, analyze its relationship with CD147. To study the expression of ΔNP63/ TAP63 in nomal skin and eyelid BCC , in order to reveal its molecule mechanism in tumorigenesis and epidermis alternation.Methods: The expression of CD147 was detected in eyelid BCC of 25 patients by fluorescent quantitation PCR and laser scanning confocal microscope immunofluorescence, the correlation between its expression and tumour recurrence、histopathology grouping were also studied. In order to analyze the relationship of MMP2 production in eyelid BCC tumour cell and matrix cell with the expression of CD147, the protein expression of MMP2 was detected by immunohistochemistry. The expression of A NP63/ TAP63 in eyelid BCC was detected by fluorescent quantitation PCR, and in different cell of normal skin fluorescent quantitation nest PCR was used. Results: (1)The RNA transcription of CD147 was detected in 25 eyelid BCC cases and only in 6 normal skin, and correspondence copy number of normalskin were more lower than eyelid BCC cases. The immunofluorescence confirmed this result. (2) The positive expression rate of MMP2 in differentiation group ^ primary undifferentiation group and recurrence undifferentiation group was separately: 40%> 47.06% and 100%, and the correlation between MMP2 and tumour recurrence was found. (3) Significant correlation was found between CD 147 expression and matrix cell, no correlation was found between CD 147 expression and eyelid BCC tumour cell. (4) The expression of A NP63 was higher than TAP63 in eyelid BCC and normal skin, and the correspondence copy number of A NP63 in eyelid BCC was significant higher than normal skin. (5) A NP63 was mainly expressed in basal layer cell and hair follicle stem cell, terminally differentiated cells almost negtive for ANP63. There was no statistical significance of TAP63 between basal layer celK hair follicle stem cell and matrix cell.Conclusion: The expression of MMP2 in eyelid BCC tumour cell and matrix cell were correlated with tumorous invasion and recurrence. CD 147 might stimulat expression of MMP2 in matrix cell of eyelid BCC. A NP63 may play an important role in the progression of eyelid BCC. Variability expression of A NP63 in eyelid BCC indicate different biological behaviour and clinic turnover of tumour cell ,and could be the index of prognosis.
