复方猫乳散对高尿酸血症及痛风性关节炎模型的主要药效学及部份机理研究

【摘要】 本课题在中、藏医药理论的指导下,通过大量藏医药经验方的筛选,首次筛选出复方猫乳散用治高尿酸血症及痛风性关节炎,在国内外尚属首次,目前已上市的药物品种以及在研项目与本处方无雷同。因而在该方的组方上具有独创性。 本课题首次对该方进行了主要药效学实验研究。实验结果发现:①该方可显著降低高尿酸血症模型小鼠(尿酸酶抑制剂造模法和次黄嘌呤造模法)的血尿酸水平。②该方可显著抑制痛风性关节炎大鼠的足肿胀;明显改善大鼠痛风性关节炎模型。③该方可显著减少醋酸致痛小鼠的扭体次数和升高热致痛小鼠的痛阈。④该方对鸡蛋清所致大鼠足肿胀、二甲苯所致小鼠耳肿胀有良好的抑制作用;但对棉球所致小鼠肉芽肿的抑制作用不明显。以上结果说明该方具有良好的降低高尿酸血症、抗痛风、抗炎、镇痛作用。其抗炎作用主要是针对急性炎症作用较好,对慢性增殖型炎症无明显疗效。以上药效学研究结果亦证明了该方用治高尿酸血症和痛风性关节炎的有效性。 本课题首次对该方进行了主要药效学相关作用机理研究。实验结果发现:①该方可显著抑制高尿酸血症模型小鼠(次黄嘌呤造模法)黄嘌呤氧化酶的活性。②该方可不同程度地降低痛风大鼠模型踝关节中IL-1β、TNFα、IL-6的水平;还可升高痛风大鼠模型中枢5-HT和DA含量;降低大鼠模型外周血清中5-HT和DA含量。该方降血尿酸作用可能与其能显著抑制黄嘌呤氧化酶作用有关;改善大鼠痛风性关节炎模型可能与其降低局部踝关节组织中细胞因子IL-1β、TNFα、IL-6的含量,调节体内单胺类

【Abstract】 The compound Maoru powder was choosed to treat hyperuricaemia and gouty-arthritis from a great deal of Tibetan proved recipes for the first time under the guidance of TCM and Tibetan medicine theory. It was original for there were no similar drugs in the market.This research concluded of chief pharmacology experiments and mechanism study. The results showed:1. It could reduce the uric acid of hyperuricaemia animal model and inhibit xanthenes oxidation enzyme(XOD).2. It could markedly relieve the swollen foot of animal model of gouty-arthritis, reduce the content of TNF-α、 IL-1β 、 IL-6 in ankle joint, increase the content of 5-HT and DA in nerve centre and reduce the content of 5-HT and DA in blood serum.3. It could remarkably decrease the twist-reaction stimulated by acetic acid, enhance the pain threshold induced by heat irritation.4. It could restrain the inflammation induced by egg white, dimethylbenzen, and tampon. All the results indicated that it had good effects on gouty arthritis and hyperuricaemia. Its treatment of hyperuricaemia had something to do with depressing XOD. The mechanism of anti-gout was decreasing the content of TNF-α 、 IL-1β 、 IL-6 in ankle joint. Its effects of abirritation and anti-inflammation was owing to adjusting the content of 5-HT and DA.Maoru and Ruxiang are discovered for the first time that it has good effect of lowering uric acid, which is original found of its function. With its further clinical application, it could developed into new drug to cure hyperuricaemia and gouty-arthritis and had academic value for this kind of Chinese medicine to develop their function theory.Taking apart experiment of Maoru Powder was done for the first time to find more effective drugs or more effective combinations.
