苏木乙酸乙酯提取物对大鼠同种异位心脏移植急性排斥反应心肌FasL mRNA表达的影响

【摘要】 心脏移植是治疗终末期心脏疾病唯一有效的手段。但是如何有效地防治急性排斥反应的发生,是关系到心脏移植成败的关键问题之一。近年来的研究资料表明,以Fas/FasL为基础介导细胞调亡的机制参与了细胞毒细胞对靶细胞的杀伤过程。 本研究以清洁级近交封闭群SD大鼠为受者,Wistar大鼠为供者,各24只,建立大鼠腹腔异位心脏移植模型。分别设模型对照组、环孢素A组和苏木乙酸乙酯提取物组,术后分期采集移植心脏。采用逆转录-聚合酶链反应和光镜技术测定心肌组织FasL mRNA的表达和病理学改变情况。 实验结果显示,苏木乙酸乙酯提取物可以明显减轻急性排斥反应对心肌的病理损伤,抑制FasL mRNA的表达。与模型对照组相比具有显著性差异。 实验结果表明,苏木乙酸乙酯提取物具有一定的抗免疫排斥作用,可以抑制急性排斥反应的发生和发展,从而对移植心脏起到保护作用。

【Abstract】 Heart transplantion has been used in many hospitals, which has become the most effective therapy for end-stage Heart disease. At the same time ,if or not the acute graft rejection is treated effectively that will become the key of heart transplating. Some study indicate recently: the Fas/Fasl is the foundation of conducting cell apoptosis, which joint the produre of CTL impairing the target cell.In this study, Adult heart clear inbred strain closed colony SD rat is the receiver, wistar rat is the provider, the sum is 24 respectively. Setting the rat cardiac allograft model.groupA: model, groupB: CsA, and groupC: sppan acetic ether extract. We will harvest heart by stage after operating, and measure the expression of histopathological change, expression of FasL mRNA by methods of microscope and RT-PCR.The result show: Sppan acetic ether extract may alleviate the impairing of acute rejection and suppress the express of fasL mRNA, there were significantly statistical differences compared with groupA.The result indicate: sppan acetic ether extract has the power of suppressing the progress of acute graft rejection. So it can provide significant protective effects on transplated heart.
