广西巴马县壮族长寿老人HLA-DQ基因的多态性研究

【摘要】 广西巴马因长寿而闻名于世,其百岁寿星率约为30.8/10万。为探讨人类白细胞抗原(HLA)与长寿的关系,我们联合应用聚合酶链式反应-序列特异引物(PCR-SSP)、寡核苷酸探针斑点杂交及DNA直接银染测序等技术对巴马县109名90岁以上长寿老人(90—103岁)、56名8旬老人(80—89岁)及71名当地健康、无血缘关系、随机抽样的成年人(23—70岁)进行HLA-DQA1、DQB1基因分型,受检者均为世居巴马8代以上的壮族人。结果如下:长寿组与对照组比较,长寿组HLA-DQA10102明显高于对照组(P=0.0026);长寿组HLA-DQA-10101及DQB10301明显低于对照组(P分别为0.0001及0.0249);长寿组与8旬组比较,各相应等位基因之间的频率无显著差异。以上结果提示,HLA-DQA10102与长寿高度关联,(OR为2.002);而HLA-DQA10101及DQB10301与长寿呈负关联,(OR分别为0.2077及0.5007)。在巴马壮族人群中,HLA-DQ基因的这3个等位基因可能直接或间接影响人类的衰老及寿命,其具体的生物学意义值得进一步研究。

【Abstract】 Bama, a remote and mountainous county in northwestern Guangxi. province in China, is well known for her Longevity: the population rate of centenarians there is about 30.8/100,000. To investigate a possible association between HLA antigens and longevity, we have geno typed the HLA-DQ A, ,DQB, loci with PCR-SSP method , dot-blot hybridization and DNA direct silver sequencing technique for 109 Longevous people (90~103yrs), 56 octogenarians( 80~89yrs) and 71 healthy, unrelated, randomly selected subjects(23-70yrs) who live in the same vicinity. All individuals tested are Bama Zhuang nationality for more than 8 generations. RESULTS: The frequency of HLA-DQA,*0102 allele was significantly high in longevous group, as compared to that of control group (p=0.0026). While the frequency of HLA-DQ A, *0101 and DQB1 *0301 of the longevous group was significantly lower than that of the control subjects (P=0.0001 and 0.0249, respectively ). No changes were found between octogenarians and longevous group.Our data suggest that HLA-DQA,*0102, DQA,*0101 and DQB^OSOl may involve in human longevity (OR=2.002, 0.2077 and 0.5007 respectively) in Bama Zhuang population. Further studies are required to clarify the biological meaning of the results.
