A seperative technology that consists of macropore resin adsorption and concentration gradient desorption was proposed. The results showed that when velocity of feed flow was 2.0 mL/min, the height and diameter proportion of column was 10∶1, operation temperature was 30℃, the yield of Puerarin could reach 93.75%.During the experiment, the thin layer chromatography was the qualitative determination; the ultraviolet derivative spectrophotometric method was the quantitative determination.
