根据已知植物病原真菌蛋白激酶 A 调节亚基基因 PKA-R(Protein Kinase A regulatory subunit)保守区域设计简并引物,通过 PCR 扩增获得了玉米大斑病菌 PKA-R 基因的同源片段。利用 Genome Walking 技术获得了 PKA-R 基因的 DNA 全长序列。利用 BLASTP 软件对该序列进行分析,结果表明该序列与小麦黄斑叶枯病菌、小麦颖枯病菌、灰霉病菌、稻瘟病菌、油菜菌核病菌的 PKA-R 基因同源性达59%~88%。Southern 杂交结果表明,PKA-R 基因在玉米大斑病菌基因组中以单拷贝形式存在。

Protein kinase A regulatory subunit gene (PKA-R) of Setosphaeria turcica was obtained by polymerase chain reaction (PCR) amplification from degenerated primer sets designed on the basis of the conserved nucleotide sequence of other fungal PKA-R gene. Then its flanking regions were obtained by Genome Walking.
