本研究以十年生露天和三年生盆栽‘曙光’油桃(Prunus persica var. nectarine cv. Shuguang.)花芽为试材,主要研究了桃树花芽RNA提取、碳水化合物代谢及其相关基因的季节性表达及低温处理下相关基因的表达变化;利用抑制差减杂交技术(SSH),以休眠期花芽RNA为Tester,以休眠解除期花芽RNA为Driver,构建花芽休眠相关基因的差减cDNA文库。主要结论如下:
1、经改良CTAB法提取的油桃花芽RNA,电泳检测条带清晰,28S与18S亮度比约为2:1,说明RNA完整性较好;核酸分光光度计检测A_(260)/A_(280)为2.0左右,说明无蛋白等杂质残留,能满足基因表达分析等后续试验的要求;而Trizol试剂盒法所提取的RNA,几乎检测不到条带,难以达到进一步试验的要求。
2、休眠期间可溶性糖(主要是蔗糖)含量尤其是在内休眠后期显著增加,淀粉含量则呈相反趋势,在内休眠后期含量降低,预示着内休眠的结束;葡萄糖和果糖含量稍有降低,变化不大;糖代谢相关基因AGPase、HisH3、HK1、SuSy、UGPase表达明显不同:AGPase无明显变化;HisH...
