本研究主要建立了一种可以同时鉴定食品中榛子及其主要致敏成分的多重PCR方法。本研究所鉴定的成分包括榛子胞浆磷酸葡糖异构酶(cytosolic phosphoglucose isomerase)、榛子致敏蛋白Cor a 1和榛子致敏蛋白Cor a 8三种蛋白。使用不同的裂解液对样品进行处理,通过对提取结果的比较,确定了适用于提取坚果中DNA成分的最佳方法;分别针对三种待测蛋白,运用专业引物设计软件Primer3和DNAstar设计出相应的引物,通过参考PCR扩增效果及NCBI的比对结果,选出特异性较强的引物对作为备选引物;进一步通过多重PCR法加以筛选,最终确定适用于多重PCR的三对引物并通过测序对PCR扩增结果进行确证。通过琼脂糖凝胶电泳和凝胶成像系统对多重PCR的扩增结果进行分析。结果表明本方法具有良好的特异性,检出限可达10 ng/μL,且反应体系各组分未出现交叉干扰,因而,本方法适合于同时检测食品中的榛子成分及榛子的主要致敏原。

This research focused on the establishment of a multiplex PCR to detect the components of hazelnut and its major allergens. The three components detected by this research were cytosolic phosphoglucose isomerase, Cor a 1 and Cor a 8. Different lysates were used to schizolysis samples.
