本试验以十五种不同籽粒颜色类型的糜子壳粉为试材,采用单因素试验及响应曲面法探讨了糜子壳粉的实验室多酚类物质提取方法;并比较了不同籽粒颜色糜子品种壳粉中的多酚类物质含量、抗氧化活性(DPPH法、β-胡萝卜素-亚油酸乳化液法和ABTS法)及酚酸类物质和黄酮类物质的组成。研究结果如下:
(1)糜子壳中多酚类物质提取的最佳工艺条件为丙酮浓度90%、料液比1:30(g/mL)、提取次数5次。在最佳工艺条件下多酚得率达到1.46%。
(2)糜子品种籽粒颜色越浅,其壳粉的L*值也就越高;糜子颗粒颜色越接近红色,其a*值就越大。糜子壳粉中的总酚含量为935.73~1909.29mg GA eq/100gDW,是小麦麸皮中多酚含量的3~6倍,且主要以结合酚形式存在。糜子籽粒颜色越深,其中的自由酚含量和自由黄酮含量也越高;糜子壳粉的总黄酮含量的变化范围为24.79~68.24 mg Rutin eq/100gDW壳粉;且主要以游离黄酮形式存在,糜子外壳颜色越深,其自由酚含量与自由黄酮含量越高;不同颜色糜子壳粉自由酚提取物的DPPH·清除能力、ABTS·清除能力、AAC值的顺序是:褐色品种>红...
