猪链球菌2型(Streptococcus suis type 2, SS2)是重要的人畜共患病病原体,主要感染人和仔猪,典型症状是脑膜炎、肺炎、败血症及突发性死亡。SS2细胞壁蛋白质有30余种,39KD细胞壁蛋白是其中之一。本试验对39KD细胞壁蛋白的黏附作用和细胞毒性作用分别进行研究。试验包括:第一,提取SS2的胞壁蛋白,利用分子筛层析法纯化39KD细胞壁蛋白至电泳纯;第二,膜上细胞黏附筛选试验和细胞黏附抑制试验。将胞壁蛋白进行SDS-PAGE电泳,并电转移至PVDF膜,用生物素标记的HEp-2细胞与PVDF膜上的蛋白条带进行黏附,用亲和素-HRP-DAB显色,得39KD阳性带。为进一步证实39KD细胞壁蛋白的黏附功能,将5μg/mL 39KD细胞壁蛋白与HEp-2细胞37℃,孵育2h,再加入SS2菌悬液(1×10~8CFU/mL) 37℃孵育2h,进行了黏附抑制试验,结果SS2对HEp-2细胞的黏附水平显著降低,提示39KD细胞壁蛋白可以部分阻断SS2对HEp-2细胞的黏附。第三,39KD细胞壁蛋白的细胞毒性试验。将5μg/mL 39KD细胞壁蛋白与HEp-2细胞37℃,孵育12 h,光学显...

Streptococcus suis type 2 (SS2) is an important zoonotic pathogen, which causes meningitis, pneumonia, septicemia and even death of both human beings and pigs.
