大白菜(Brassic campesstris L ssp. pekinensis)属于二年生蔬菜作物,需要经历一个低温阶段才能顺利完成发育转变。大白菜为种子春化型植物,萌动的种子即可感受低温完成春化,进而抽薹开花。培育适宜种植、具有强冬性的耐抽薹品种,已成为亟待解决的问题。
大白菜通过春化到抽薹开花其间表现出一叶色转变过程,叶色转变后就会现蕾进而抽薹开花,在弱冬性品种上的表现尤为突出。本研究以不同冬性的大白菜品种为材料,从激素、色素含量变化和蛋白质组分变化研究大白菜抽薹过程发生叶色转变的生理机制,同时利用拟南芥基因同源序列克隆大白菜AP1基因片段,运用RT—PCR进行不同冬性品种叶色转变过程中AP1基因表达分析,探讨叶色转变可能分子机制。主要结果如下:
1.不同冬性的大白菜品种抽薹过程中色素含量变化不同,其中强冬性品种叶绿素a、叶绿素b、类胡萝卜素的峰值均出现在花茎分化期,最低值出现在花芽分化期。而中等、弱冬性品种三种色素的峰值均出现在叶色转变后的花茎伸长期,最低值出现在花茎分化期。抽薹过程中叶绿素(a/b)的比值在叶色转变期为最高。
