本试验以蒙古冰草( A.mongolicum Keng )、诺丹冰草( A.desertorum cv.‘Nordan’)和蒙农杂种冰草(A.cristatum×A.desertorum cv.‘Mengnong’)为材料,以幼胚为外植体,研究胚龄、基因型、供试培养基和4℃低温预处理时间对冰草愈伤组织诱导的影响;针对诺丹冰草和蒙农杂种冰草初生愈伤组织质量较差这一情况,研究了它们的最佳继代培养基;同时研究了培养基、基因型、激素配比和活性炭浓度对冰草愈伤组织分化的影响。试验结果表明:
1)蒙古冰草愈伤组织诱导培养基:MS+CH(500mg/L)+2,4-D(2.0mg/L)+6-BA (0.05mg/L);继代培养基:MS+CH(500mg/L)+2,4-D(1.0mg/L);分化培养基:MN+CH(500mg/L)+NAA(0.5mg/L)+6-BA(4.0mg/L)
