目的:探讨赤芍总甙(Total glucosides of Radix Paeoniae Rubra,TGC)作用于S_(180)肉瘤小鼠,较系统分析TGC诱导肿瘤细胞凋亡和细胞因子分泌的影响,进一步完善TGC的抗肿瘤作用机制。方法:瘤鼠造模后,采用赤芍总甙灌胃7天后,测定指标。1抑瘤率:将各组剥离完整的肿瘤组织称重,计算其数值;2 病理学观察:光镜、电镜观察各组肿瘤组织病理学特点;3 原位杂交法观察TGC对小鼠c-myc癌基因mRNA转录的影响;4 通过SABC免疫组化法,观察TGC对小鼠Bcl-2、P16基因蛋白表达结果;5 ELISA法测定IL-10,观察TGC对肿瘤细胞细胞因子的影响。结果:TGC组、TGC+CY组、CY组与模型组的抑瘤率表达显著性差异。肿瘤组织病理学观察显示,赤芍总甙具有明显抑制肿瘤作用。TGC组与模型组Bcl-2、c-myc阳性表达有显著性差异,Bcl-2抑制细胞凋亡作用;c-myc是原癌基因,TGC组Bcl-2、c-myc表达呈下降趋势。P16为抑癌基因,TGC组表达增高。赤芍总甙能降低肿瘤细胞IL-10的分泌,抑制肿瘤细胞的增殖。结论:赤芍总甙具有调节细胞因子,纠正荷瘤机体的T...
