从 2002 年 8 月至 2003 年 9 月,从云南省主要烟草种植区采集新鲜烟
草植株用于分离内生真菌。用组织分离与表面消毒有效性评价相结合的方
法确定了烟草内生真菌的有效分离方法及相关技术参数。次氯酸钠消毒是
关键步骤,应根据不同生长期的不同烟草组织选择适宜的消毒时间,在保
证表面消毒效果的前提下获得尽可能多的菌株。
获得的 186 个菌株在通常条件下多不产孢,经过多种方法的诱导产
孢,主要是近紫外线照射和培养基中添加烟草组织,共有 70 个菌株在人
工培养条件下产孢,鉴定为 10 个种,分别是:烟草黑痣菌(Phyllachora
nicotiae W. Han sp. nov.),烟生黑痣菌(Phyllachora nicoticola W. Han sp.
