A new SDS-PAGE method of high molecular weight (HMW) subunit of wheat glutenin was found, through reference to the method of protein SDS-PAGE by B. J. Smith (1984). The new method can analyse a full-single grain or a semisingle one. According to the electrophoretograms of 5071 full-single wheat varieties and more than 1600 semi-single hybrid progenies, it has been proved to have characters of high resolving power, appropriate gel strength, simple and rapid to use and bands in dry slab without spreading.
