桔梗为桔梗科(Campanulaceae)植物桔梗Platycodon grandiflorum (Jacq) A. DC.的干燥根。本课题应用现代色谱分离技术和免疫学方法,以溶血性与免疫佐剂活性评价为导向,从中药桔梗中分离出4个佐剂活性皂苷,采用多种光谱技术确定了化学结构。通过检测这4个皂苷的溶血性及对模式抗原卵清蛋白(OVA)的佐剂活性,探讨了桔梗酸类皂苷溶血性或免疫佐剂活性与化学结构之间的构效关系;评价了桔梗皂苷D (PlatycodinD, PD)和桔梗皂苷D2 (PlatycodinD2, PD2)对重组乙型肝炎表面抗原(recombinant hepatitis B surface antigen, rHBsAg)疫苗的佐剂效应和特点,以及Platycodin D对商品禽流感新城疫重组二联活疫苗的佐剂作用。
