The Experimental Study of the Effects on Rat Kidney Caused by Different Dosage Forms of Aristolochia Debilis Sieb.Et Zucc and Complex Prescription Contains Aristolochia Debilis Sieb.Et Zucc
目的:通过动物实验观察青木香的不同剂型及含青木香复方即青木香汤剂、青木香散剂、冠心苏合丸剂对大鼠肾脏的影响,研究青木香剂型对其肾毒性作用的影响;测定青木香汤剂、青木香散剂、冠心苏合丸剂中马兜铃酸A的含量,以研究青木香中马兜铃酸A的煎出量与药物剂型的关系。
方法:健康wistar大鼠80(200±10g,♀∶♂=1∶1),随机分为4组,每组20只,分别为青木香汤剂组、青木香散剂组、冠心苏合丸组、对照组。分别给予青木香汤剂:0.81g·kg~(-1)·d~(-1)灌胃、青木香散剂:0.81g·kg~(-1)·d~(-1)、冠心苏合丸:0.32g·kg~(-1)·d~(-1),对照组给予等体积蒸馏水灌胃,每日1次,连续3个月。实验期间每周称1次体重,根据体重调整给药剂量。于第1、2、3月末检测大鼠BUN、SCr、尿β_2-MG、尿NAG;于第3月末取肾组织,通过光镜和电镜观察肾脏组织病理学变化;免疫组织化学方法检测肾组织TGF-β1、CTGF的表达;real time PCR检测肾组织CTGF mRNA的表达,HPLC法检测肾组织马兜铃酸A的含量。并用HPLC法检测青木香散剂、冠心苏合丸、青木香汤...
