本文以当年及多年生平邑甜茶(Malus hupehensis (Pamp) Rehd. var pinyiensis Jiang)为试材,采用水培及土壤盆栽试验相结合的方式,研究了根系精氨酸代谢与一氧化氮(NO)和多胺产生的关系、NO的产生途径,以及NO对平邑甜茶根系生长发育和抗逆性的影响,结果如下:
1.应用Luminol-H2O2化学发光法测定植物NO含量和一氧化氮合酶(NOS)活性,通过添加L-精氨酸、NOS抑制剂、NO吸收剂、酶液煮沸杀酶等证明该方法是一种有效的测定植物NO含量和NOS活性的方法。
2.平邑甜茶幼树根内精氨酸含量显著高于茎和叶片;根内精氨酸含量以多年生根最高,叶片完全展开后其含量达到最高,后随地上部生长逐渐下降。平邑甜茶幼树不同器官NO含量及NO生成相关酶活性有很大差异。NO含量、NOS和NI-NOR活性根中最高,其次为嫩茎,叶片中最低。细根和嫩茎线粒体具有以NADH为电子供体还原亚硝酸盐生成NO的能力,而叶片线粒体不具备这种途径。多胺途径和NO途径的变化具有一致性,与组织内精氨酸含量呈正比例关系,且具有组织特异性差异,细根较叶片具有较...
