灰树花[ Grifola frondosa (Dicks. Fr.) S. F. Gray ]是近年来开发比较多的珍奇药食用菌之一,其多糖和多糖复合物在抗肿瘤、抗病毒、增强免疫、降血糖、治疗糖尿病等多方面的具有明显的生物活性。本论文在实验室前期研究工作的基础上,对灰树花菌丝体的深层发酵培养基优化、扩大培养、菌丝体中抑瘤活性物质的分离和筛选、组成结构、抑瘤活性物质的体内外抑癌活性及机理等方面进行了较为深入的研究。根据实验室前期工作所得到的结论,利用Box-Behnken 3×3设计优化了影响灰树花深层发酵菌丝体和胞外聚合物的生产的主要培养基组成碳源(葡萄糖)、氮源(蛋白胨)和无机盐(KH_2PO_4)。响应面分析结果表明,各因素及其交互作用对响应值(灰树花菌丝体和胞外聚合物产量)均存在显著的相关性。通过典型性分析,获得最大菌丝体(17.61 g/l)时培养基组成为:葡萄糖45.2 g/L,KH_2PO_4 2.97 g/L,蛋白胨6.58 g/L,MgSO_4·7H_2O 1 g/L和玉米浆15 g/L;而获得最大胞外聚合物量(1.326 g/L)时的培养基组成为:葡萄糖58.6 g/L...
