龙须菜(Gracilaria lemaneiformis)是一重要的产琼胶红藻,也是江蓠属的一个重要养殖物种。同时龙须菜还是一种很好的遗传学和分子生物学研究对象。我们以龙须菜为主要材料,进行了cDNA文库的构建、表达序列标签(EST)的分析,以及简单序列重复区间扩增多态性(ISSR)的分析。
在龙须菜配子体的cDNA文库构建中,使用UNIQ-10 Trizol Total RNA Preparation Kit来进行总RNA的提取,将cDNA Synthesis Kit和PCR cDNA Library Kit组合使用进行cDNA的扩增,最后将cDNA与pMD 18-T载体连接后转化到大肠杆菌JM109中。并从该文库中随机挑取克隆进行EST的分析。为了研究不同世代、不同性别基因表达的差异,我们又构建了龙须菜四分孢子体的cDNA文库。用RNeasy Plant Mini Kit进行总RNA的提取;用SMART cDNA Library Construction Kit进行cDNA的合成与文库的构建,包装系统购自Promega公司。结果显示该cDNA文库中含有1....

Gracilaria lemaneiformis is one of important agarophytic species of red alga and important cultivable species in the genus. G. lemaneiformis is also an ideal material for studies of genetics and molecular biology. The cDNA libraries of G. lemaneiformis have been constructed, and analyses of expressed sequence tags (EST) and inter-simple sequence repeat (TSSR) have also been carried out.
