Pharmacokinetics Study of Multiple Components of Traditional Chinese Medicine in Rat Plasma by Ultra Performance Liquid Chromatography Tandem Mass Spectrometry
中药的成分复杂,治疗的效果是多种成分相互作用的结果,因此,研究中药的活性成分在生物体内的动态变化过程及规律,阐明药物吸收、分布的药代动力学特征十分重要。超高效液相色谱串联质谱技术具有选择性强、灵敏度高等特点,在中药的药代动力学研究应用越来越广泛,是研究药物代谢的有效手段。本论文应用超高效液相色谱串联质谱技术,分别测定大鼠血浆中蒽贝素及中药材三白草、紫花地丁多种活性成分的含量,并建立相应的血浆浓度-时间曲线,计算药代动力学参数,探讨药代动力学特征,为促进中药开发、阐明中药作用机制和设计合理给药方案提供科学依据。1.应用超高效液相色谱串联质谱法同时测定三白草中原儿茶酸、芦丁、金丝桃苷、异槲皮苷、槲皮苷、木犀草素和三白草酮7种生物活性成分的含量,并应用于在大鼠体内的药代动力学研究。血浆样品通过蛋白沉淀方式处理,以葛根素和大豆苷元为内标,使用含有0.10%乙酸水和乙腈进行梯度洗脱,采用多反应监测模式进行检测。原儿茶酸、芦丁、金丝桃苷、异槲皮苷、槲皮苷、木犀草素和三白草酮在大鼠血浆中的线性范围分别为 15.0-600 ng mL-1、25.0-400ng·mL-1、15.0-600 ng mL-1、15.0-300 n...
