茶树是我国重要的经济作物之一。近年来,已育成许多具有优异性状的茶树品种。一方面,这些品种大多采用系统选种和杂交育种的方法选育而成,骨干亲本类似,亲缘关系比较接近,使品种的遗传背景较窄。如果能从分子水平上揭示这些品种之间的亲缘关系和遗传结构,将为杂交育种时亲本的选择提供一定的科学指导。另一方面,随着,新培育出的优良品种越来越多,对茶树品种的鉴别提出了更高的要求。SSR(Simple sequence repeat,简单重复序列)分子指纹图谱鉴定技术具有简便、迅速不受环境条件影响等优点,非常适合品种鉴定。本研究以254个优良茶树品种为材料,从前期构建的遗传连锁图谱上选出185个SSR标记,分析了我国茶树品种的遗传多样性水平,并通过筛选出的核心引物,构建了较全面的茶树品种分子指纹图谱。主要研究结果如下:1.从185个SSR标记中初步筛选出了128个扩增条带清晰、稳定的SSR标记。不同连锁群上的SSR标记检测到的平均等位基因数、基因型个数、多态性息含量均有较大差异。根据引物扩增条带易统计程度,PIC(Polymorphism information content,多态性息含量)和基因型个数,筛选了2套均匀分布于15...
