目的:使用重组人干扰素α-2b对大鼠慢性坐骨神经缩窄性损伤模型(chronic constriction injury of the sciatic nerve,CCI)行椎间孔注射,通过测定热痛阈值、脊髓和背根神经节中p-p38MAPK含量,观察其是否对CCI诱发的神经病理性疼痛有治疗作用,并探讨分析其可能机制。方法:实验分为四组:假手术组、CCI组、6万单位组和8万单位组,每组各12只,共48只,均为随机选择的雄性大鼠。假手术组在CCI造模时,仅切开暴露坐骨神经,不进行结扎。其它三组暴露坐骨神经后,用丝线行间隔1mm的4道结扎。术前1天、术后1天、3天、7天分别测定各组热痛阈值,确定造模成功。第8天,对除假手术组外的其它三组手术侧分别行椎间孔注射,CCI组给与0.1ml生理盐水,6万单位组给与0.1ml6万单位重组人干扰素α-2b,8万单位组给与0.1ml8万单位重1组人干扰素α-2b,假手术组不作处理。各组于阻滞后的1天、3天,分别进行热痛阈测定。于椎间孔注射的前1天,阻滞后1天和阻滞后3天,各组随机取4只大鼠,取L4-5段的脊髓和手术侧的背根神经节,裂解处理后,用酶联免疫吸附测定法(elisa)法测定...
