天然橡胶是世界上四大战略物资之一,在我国的国家安全和国民经济中发挥着重要作用。由多主棒孢(Corynespora cassiicola)引起的橡胶树棒孢霉落叶病(Corynespora leaf fall disease, CLFD)现已成为南亚、东南亚和中非橡胶树最具破坏性的叶部病害之一,是继南美叶疫病(South American Leaf Blight, S ALB)之后第二大威胁世界天然橡胶产业的重要病害。国内外对多主棒孢病菌分子致病机理的研究报道极少。本研究拟通过限制性内切酶介导转化(Restriction enzyme-mediated integration, REMI)体系的优化,获得橡胶树多主棒孢病菌遗传转化子库,并对其进行质量评价,转化子表型观察和致病性缺陷型转化子筛选。同时,选取强致病力菌株Cc01,与北京六合华大基因科技股份有限公司合作进行该菌株全基因组序列测定;并采用同源克隆法从多主棒孢Cc01中克隆hog1基因,对其功能进行初步分析。获得以下研究结果:利用限制性内切酶介导转化方法,采用KpnⅠ、HindⅢ、 SacⅠ和SmaⅠ线性化的质粒pUCATPH转化橡胶树多主棒孢Cc01菌株...
