目的:1.通过离体培养大鼠星形胶质细胞,观察P物质对星形胶质细胞的活化作用,包括细胞形态学观察和对IL-1β和IL-10释放量的检测。2.通过P物质作用于离体培养的星形胶质细胞,观察星形胶质细胞胞内Ca2+浓度的变化,探讨活化可能的机制。方法:1.取出生24 h以内的SD乳鼠原代培养星形胶质细胞。将培养的星形胶质细胞分为5组:(1)对照组,细胞孵育全培液;(2)全培液+P物质30 n M组;(3)全培液+P物质100 n M组;(4)全培液+P物质300 n M组;(5)全培液+P物质1000 n M组。各组细胞分别在孵育2 h、6 h、12 h、24 h后收集细胞上清液同时保留细胞,以便进行后续实验。2.采用免疫细胞荧光法鉴定GFAP阳性的星形胶质细胞,并观察P物质孵育后星形胶质细胞在各浓度以及各时间点的形态变化。3.采用ELISA的方法检测P物质孵育后星形胶质细胞在各浓度及各时间点释放IL-1β和IL-10的情况。4.利用比例高内涵成像系统的胞内钙离子成像技术观察P物质作用于星形胶质细胞后,其胞内Ca2+浓度的变化。结果:1.培养的星形胶质细胞的鉴定:经免疫细胞荧光法鉴定,培养的细胞为GFAP阳性的星形胶...
