中国传统医药源远流长,博大精深,采用现代生物技术手段研究阐述传统中药的抗肿瘤机理对中药的现代化具有非常重要的意义。多年临床经验表明传统中药白花蛇舌草有清热解毒、活血止痛及利尿消肿等多重功效,同时其对炎症和肿瘤也具有显著的疗效作用。近年来研究发现传统中药白花蛇舌草提取物能够诱导多种肿瘤细胞凋亡,但目前未见与其抗肿瘤作用机理相关的研究报道。真核细胞转录激活因子3(Signal Transducer and Activators of Transcription 3,STAT3)及其参与的相关信号通路与肿瘤细胞抗凋亡、增殖、入侵、迁移和血管生成等肿瘤发生发展过程密切相关,其已成为多种恶性肿瘤治疗与靶点抗癌药研发的焦点。本研究旨在以STAT3信号通路为靶点评价传统中药白花蛇舌草提取物在细胞水平及动物体内的抗肿瘤活性并探讨其作用机理。首先采用MTT细胞增殖实验初步评价了中药白花蛇舌草总提取物(HT)、乙酸乙酯相提取物(HE)和水相提取物(HW)三组分在细胞水平上的抗肿瘤活性,结果表明HE组分能够显著抑制人乳腺癌细胞MDA-MB-468及人前列腺癌细胞DU145的生长。免疫印迹实验结果进一步表明HE组分能够特异性的抑制S...
