本文首先以‘哲引3号杨’(Populus pseudo-simonii×P. nigra 'Zheyin3#')为母本,‘北京杨’(P.×beijingensis)为父本,在通过杂交获得‘哲引3号杨’ב北京杨’杂种种子的基础上,建立起含有200个基因型的杂种全同胞家系的组织培养体系;进而开展了多基因型青杨杂种叶片离体再生研究,首次探讨了基因型、基因型×培养基互作效应对青杨叶片离体再生的影响;并依托建立的多基因型青杨杂种叶片离体再生技术,展开了秋水仙碱诱导青杨离体叶片体细胞染色体加倍研究,主要研究结果如下:
1.建立了‘哲引3号杨’ב北京杨’多基因型叶片不定芽诱导再生技术体系,为后续的离体叶片体细胞染色体加倍打下了基础。
首先通过完全随机区组试验设计,研究3种植物生长调节剂、7种培养基对‘哲引3号杨’ב北京杨’全同胞家系内5个基因型叶片离体再生的影响,初步筛选出适合5个基因型青杨杂种叶片离体再生的培养基(Fs) MS+6-BA0.4mg·L-1+NAA0.05mg·L-1.在此培养基上,叶片培养10d左右切口处形成突起,20d时形成完整的不定芽,分化率达到59.3%。
