【目的】研究实验性光损伤小鼠视网膜色素上皮细胞(retinal pigment epithelium,RPE)中金属蛋白酶组织抑制物-1(tissue inhibitor of metalloproteinase-1,TIMP-1)表达的变化,以及促红细胞生成素(erythropoietin,EPO)对其的影响,探讨EPO对视网膜光损伤发挥保护作用的机制。【方法】52只健康成年近交系BALB/C小鼠,随机分为单纯光照组(24只)、EPO处理组(24只)和正常对照组(4只)3组。其中前两组按光照后处死时间的不同又各自分出6h、12h、36h、72h、96h和7d6个亚组,置于6000LUX光照箱中照射4h,EPO处理组小鼠光照前2小时腹腔注射重组人促红细胞生成素(recombination human erythropoietin,rHuEPO),剂量为5000U/kg。采用苏木素-伊红(HE)染色观察RPE细胞形态学变化、免疫组织化学法检测其TIMP-1、细胞外信号调节激酶(ERK-1)蛋白的表达。【结果】(1)形态学发现:正常小鼠视网膜RPE细胞呈单线状排列,细胞形态规整。单纯光照后12...
